A Palestinian-Jordanian mother and an Egyptian banker father gave birth to Calamawy on October 28, 1986, in Bahrain. Her sibling is older. She was primarily raised in Bahrain, but before she was twelve, she also spent six years bouncing around between Houston, Texas, and Doha, Qatar. Calamawy is bilingual in both languages.
She was motivated to pursue acting after seeing the 1992 movie Death Becomes Her as a young girl.
Calamawy graduated from high school in Bahrain at the age of 17, and at the insistence of her father, she travelled to Boston, Massachusetts to pursue a career in industrial design. After that, she spent five years living in Dubai before returning to the United States to pursue a career in acting.
She submitted an application to Emerson College and promised her parents she would attend if accepted. She got in and graduated with a B.A. in theater studies. Also in New York City, Calamawy attended the William Esper Studio.

May El Calamawy was the name under which Calamawy was credited when she first began appearing in short films. Later, she substituted May Calamawy. She took part in the New York Arab American Comedy Festival after finishing college. She split her time between Dubai and Abu Dhabi from 2009 to 2014 while appearing in short films and a TV pilot.
Her breakthrough performance came in Tobe Hooper’s 2013 horror movie Djinn, the first to be made in the United Arab Emirates.
She had guest starring roles on The Brave and Madam Secretary in 2017 in addition to a recurring role in the National Geographic miniseries The Long Road Home. She had a guest appearance in the CBS criminal drama television series FBI the following year.
She will play Dena Hassan, Ramy’s sister, in a recurring role in the Hulu comedy-drama Ramy, it was revealed in October 2018. She voiced Ellie Malik in the NBA 2K21 video game in 2020.
She starred in the 2021 movie Together Together alongside Ed Helms and Patti Harrison. Calamawy was cast as Layla El-Faouly, an Egyptian archeologist and Marc Spector’s estranged wife, in the Disney+ miniseries Moon Knight, which premiered on March 30, 2022.
This was revealed in January of the same year (portrayed by Oscar Isaac). The first Arab superhero and character in the Marvel Cinematic Universe was Layla.
She will appear as a guest star on Marvel’s Moon Girl and Devil Dinosaur, it was confirmed in July 2022. In 2023, the program is scheduled to debut on Disney Channel and Disney+.
